Aspergillus fumigatus Specific IgE ELISA Principle : 

The Aspergillus fumigatus IgE ELISA is based on an indirect enzyme-linked immunosorbent assay. Patient serum is added to microtitre wells pre-coated with Aspergillus fumigatus allergen. Allergen-specific IgE antibodies present in the sample bind to the coated allergen.

Following washing, HRP-conjugated anti-human IgE is added to detect bound IgE antibodies. TMB substrate is subsequently used for colour development. After stopping the reaction, absorbance is measured at 450 nm, with 630 nm used as a reference wavelength. Specific IgE concentration is determined using a calibration curve and expressed in IU/ml.
